چکیده

Recent advances in gas-liquid chromatography have permitted the separation and identification of aldehydes derived from plasmalogens, and a number of analyses of aldehydes from animal plasmalogens have been reported (l-3). Bacterial plasmalogens, on the other hand, have only recently been discovered, and no analyses of their fatty aldehydes are available (4-6). Such analyses would be of particular interest since the fatty acids of bacteria differ from those of higher organisms in a number of features, which reflect differences in biosynthetic pathways (7,8). These features include: the presence of cis-vaccenic acid instead of oleic acid as the major 18 carbon monounsaturated fatty acid (9), the presence in many bacterial species of fatty acids containing a cyclopropane ring (lo), and the absence of polyunsaturated fatty acids. We have undertaken an analysis of the fatty aldehyde chains of Clostridium butyricum plasmalogens. Evidence is presented for the occurrence of 16 and 18 carbon saturated and monounsaturated aldehydes, and 17 and 19 carbon aldehydes which contain a cyclopropane ring. No polyunsaturated aldehydes appear to be present.

Phospholipids of Clostridium butyricum. IV. of monounsaturated and cyclopropane fatty acids and alk-Lenyl ethers by capillary column chromatography Analysis of the positional isomers

The positional isomers of the cyclopropane fatty acids of Clostridium butyricum phospholipids have been analyzed by capillary column gas-liquid chromatography. Greater than 95% of the methylenehexadecanoic acids was the 9 , l O isomer. On the other hand, 6G70yO of the hexadecenoic acid precursors was the A7 isomer, and the remainder was the A9 isomer. Of the methyleneoctadecanoic acids 75580% w...

Formation of acyl and alkenyl glycerol derivatives in Clostridium butyricum.

The mechanism of synthesis of plasmalogens is not currently known, but the incorporation of labeled phosphate, acetate, fatty acids, and fatty aldehydes into plasmalogens has been described for intact systemsl-B. Such isotopic studies have suggested a variety of pathways leading to the synthesis of plasmalogens. Among these proposed pathways are the dehydrogenation of the alkyl ether bond, the ...

Highly enantioselective addition of phenylethynylzinc to aldehydes catalyzed by chiral cyclopropane-based amino alcohols.

The enantioselective addition of phenylethynylzinc to aldehydes catalyzed by a series of cyclopropane-based amino alcohol ligands 7 was investigated. The reactions afforded chiral propargylic alcohols in high yields (up to 96%) and with excellent enantioselectivities (up to 98% ee) under mild conditions. Furthermore, studies on the structural relationship show that the matching of the chiral ce...

Quantitative micro determination and isolation of plasmalogen aldehydes as 2,4-dinitrophenylhydrazones.

A micro spectrophotometric procedure for the quantitative determination of plasmalogen aldehydes is described which utilizes simultaneous methanolysis and formation of 2,4-dinitrophenylhydrazones. After isolation of the hydrazones by thin-layer chromatography, the aldehydes can be regenerated, reduced, acetylated, and then analyzed by gas-liquid chromatography. Identification of the plasmalogen...

Synthesis of fatty aldehydes and their cyclic acetals (new derivatives for the analysis of plasmalogens).

Saturated and unsaturated fatty aldehydes were prepared in good yield by the reduction of acid chlorides with lithium aluminum tri-t-butoxy hydride. Saturated odd and even numbered aldehydes were prepared by the ozonolysis-reduction of 1-alkenes. Ozonides were hydrogenated with a Lindlar catalyst or reduced with dimethyl sulfide. Several diols, including 1,3-propanediol and ethylene glycol, wer...
